| I. Background | |
| GRM4(代謝型谷氨酸受體 4)是一種抑制性 C 族 GPCR,通過(guò)激活 Gi/o 蛋白抑制腺 苷酸環(huán)化酶、降低胞內 cAMP 水平,并觸發(fā) PI3K/Akt 抗凋亡通路。在 中樞神經(jīng)系統 中, GRM4 高表達于小腦顆粒細胞、海馬及基底節,負調控突觸前谷氨酸/γ-氨基丁酸(GABA) 釋放,參與運動(dòng)協(xié)調(皮質(zhì)-紋狀體通路)、突觸可塑性及神經(jīng)元興奮性平衡。病理狀態(tài)下, GRM4 功能異常驅動(dòng)帕金森病多巴胺能神經(jīng)元凋亡、黑色素瘤免疫逃逸、精神分裂癥認知 缺陷及神經(jīng)病理性疼痛等。 | |
